It all began on August 1, 1926, when, at the initiative of N.F. Erna, a meeting of Russian Orthodox Christians living in Asuncion and the provinces of Paraguay was held in the apartment of Prince Y.K. Tumanov. At this meeting, it was decided that it was necessary to create an Orthodox church in Asuncion.

Metropolitan Evlogy was informed that the Russian community in Asuncion, which at that time numbered 98 people, held a meeting and elected a committee to explore the financial possibilities of inviting a priest and building at least a small church.

On September 5, 1927, part of the Russian community in Asuncion, led by the church committee, solemnly welcomed Archimandrite Pakhomiy, the first rector of the new parish. A thanksgiving service was held in the temporary church located in the former "Russian House," after which everyone gathered for tea. The first Divine Liturgy took place on September 11.

The church was built according to the design of Captain Georgy Leonidovich Shmagailov, a talented military engineer who emigrated to Paraguay. In the Russian Empire, he was involved in the construction of the Grodno Fortress, the last one before World War I.

The parish chronicle notes that 15-20 people usually attended the services. Festive services and memorials for the Emperor and his family, General Wrangel attracted almost all Orthodox Russians living in Asuncion.

In 1978, a Russian newspaper reported that Russians in Asuncion enjoyed favorable treatment from the government led by President Alfredo Stroessner. At the funeral of Colonel Andreev and Major Korsakov, the president and all ministers were present, and military units held an honor guard at the coffin.

Today, the composition of the parish has become international: among the parishioners are Russians, Ukrainians, Belarusians, Greeks, and a woman from Macedonia sings in the choir. Bulgarians and Serbs come as guests, and there are also ethnic Paraguayans.
