Initially, the hospital was located in 13 separate wooden buildings. It included 150 beds, a charity house for 100 people, a workhouse for men, and a nursing home for former military personnel. Over time, branches of the Catherine Hospital began to appear in different districts of Moscow, which later became independent medical institutions: Novo-Catherine, Myasnitskoye, Yauzskoye, Basmannoye, Preobrazhenskoye, and others.

In 1835, the hospital was renamed Old-Catherine. After the hospital organized on Strastnoy Boulevard near the Petrovsky Gates separated and was called New-Catherine, the former was named Old-Catherine.

In 1890, a maternity ward with 7 beds opened in the gynecological department of the Old-Catherine Hospital. In this ward, in 1896, Professor Dmitry Vigilev performed the first cesarean section in Moscow.

In 1906, two more branches of the maternity ward were opened.

In 1907, the maternity shelters were combined into one institution, the City Maternity Hospital at the Old-Catherine Hospital.

In 1908, with funds from the Morozov family, two floors of a new maternity hospital building were constructed.

In 1910, with donations from patrons, a three-story building with the main entrance from 3rd Meshchanskaya Street was erected, named "Maternity Hospital named after the Morozovs at the Old-Catherine Hospital." Later, its name changed: in 1924, the maternity hospital became known as the "Proletarian Maternity Hospital." Since 1936, it functioned as City Maternity Hospital No. 8 of the Dzerzhinsky District of Moscow.

The hospital changed its name several times: Moscow Clinical Institute (MKI), Moscow Regional Clinical Institute (MOKI, MOKI MedVUZ), Moscow Regional Research Clinical Institute (MONIKI).

Since 2012, it has been called "State Budgetary Healthcare Institution of the Moscow Region Moscow Regional Research Clinical Institute" (GBUZ MO MONIKI).
