The church was built in 1907 on a hill at the beginning of Church and Bazaar streets, thanks to the efforts of the entire community. The design was based on the Resurrection Church from Shadrinsk, built in 1880. The painting was done by artists from the same city. The church, named in honor of the Holy Trinity, was consecrated in 1908 on the eve of the corresponding holiday. The first priest was Father Vitaly Shmotin, and the deacon was Nikolai Gruntsevsky, transferred from the village of Varlakovo.

In November 1936, the priest of the Mishkino church, Father Vasily Nikolaevich Polovinkin, church council member T.S. Ponomarev, and church elder A.P. Ankudinov were arrested, accused of anti-Soviet and religious agitation.

In May 1937, meetings of labor collectives took place at enterprises in Mishkino, where the closure of the church was discussed. The decision to close it was made unanimously. Soon the church was closed, the bells were removed and sent for melting down, part of the fence was used for railings in the school, and the building was turned into a storage for bran.

From 1940 to 1946, the former church housed a grain warehouse of the State Insurance Fund. The Great Patriotic War changed attitudes towards the church, and on the eve of 1945, A.A. Antonova, 20 widows, and two elderly men who participated in the church's construction sent a request to the Council of Ministers of the USSR to allow the church in Mishkino to reopen.

In early March 1947, the church building was returned to the believers, and it was quickly restored. Icons for the iconostasis were brought from the homes of local residents. The church was reopened and consecrated on the eve of the Holy Trinity holiday in May 1947 by priest Mikhail Pavlovich Biryukov from Shadrinsk. At that time, the priest was Father Vasily, Vasily Vasilievich Gomzikov, who served as rector until 1989 and is buried in the churchyard. Since 1989, the rector has been Father Alexey Fasola, who has done much for the church: he acquired bells, restoring the ringing of bells in Mishkino, and updated the heating system.
