The history of Orthodoxy in Bolivia is relatively short even compared to other South American countries. Bolivia has remained unsettled and unstable for most of its independent existence, which explains why emigrants often chose other countries in the region, such as Argentina or Brazil, for relocation. There have always been few Orthodox Christians in Bolivia, and they appeared here later than in other places. The actual church presence began only in the 21st century.

The first Orthodox church in Bolivia was recently built by Russian Christians in the country's largest city, La Paz. In three years, enough funds were raised for construction thanks to about 50 donors, including employees of the Russian embassy in Bolivia. Architect Larisa Kusablik, who lives in Bolivia, prepared the church project. The iconostasis was made in Sergiev Posad, and one of the churches in New York donated ten icons. The Moscow Patriarchate provided church utensils.

On February 24, 2008, Metropolitan Platon, the ruling bishop of the Argentine and South American diocese of the Moscow Patriarchate, consecrated the church in honor of the Holy Trinity, conducted the Divine Liturgy, as well as the baptism and wedding of local parishioners.

The openness of indigenous Bolivians to Orthodoxy is confirmed by an incident that occurred on Christmas 2009, when two Bolivian citizens from the Quechua tribe were baptized in the Moscow church of St. Thomas on Kantemirovskaya. Priest Daniil Sysoev, who performed the baptism, recounted that they discussed faith, and the women read the Creed in Spanish, which he found on the internet. Their friend from Peru, belonging to the Inca tribe and living in Moscow for a long time, told them about the Orthodox faith. In baptism, the women received the names Maria and Elizabeth in honor of Mary Magdalene and the Holy Martyr Elizabeth Feodorovna Romanova. Father Daniil noted that this might be the first case in history where Quechua Indians accepted Orthodoxy.
