The village of Domnino, located in the modern-day Susanin district, holds significant historical importance for Russia as the estate of Ksenia Ivanovna Shestova, the mother of the first tsar from the Romanov dynasty, Mikhail Fyodorovich. At the end of 1612, the peasant Ivan Susanin performed his famous feat here, and according to some historians, his remains may have been buried in the basement of the wooden Resurrection Church that stood on this site.

From 1810 to 1817, a stone Assumption Church was built near the old Resurrection Church, on the site of the home of the nun Martha, with chapels dedicated to St. Nicholas and the Tikhvin Icon of the Mother of God, which were added in 1827. The wooden church, which had finally fallen into disrepair, was dismantled in 1831.

In 1879, the Alexandrovsk Orthodox Brotherhood was founded in Kostroma, which engaged in charitable and educational activities in places associated with the history of the Romanov house. In 1893, the brotherhood built a two-story stone building for a church-parish school in Domnino, and in 1913, erected a chapel in honor of Ivan Susanin in the village of Derevenki.

In the 20th century, the Assumption Church in Domnino continued to function, closing only from 1937 to 1944. Since 1991, the church has been a dependency of the Epiphany-Anastasia Women's Monastery, and the sisters of this monastery settled in the building of the former church-parish school, which was transferred to the Kostroma diocese.

According to the decision of the Holy Synod of the Russian Orthodox Church on March 25, 2004, a women's monastery was opened in Domnino in the name of the Holy Royal Martyrs: Tsar Nicholas, Tsarina Alexandra, Tsarevich Alexei, and Grand Duchesses Olga, Tatiana, Maria, and Anastasia.

The monastery includes a chapel in honor of the Beheading of St. John the Baptist, located in the village of Derevenki. A small almshouse also operates within the monastery.
