In 1862, the construction of a permanent stone church began. By 1867, a cold three-altar church with an adjoining bell tower was erected. The church was shaped like a cross with the main altar in the center and side altars. It was adorned with 16 windows and topped with a cross covered in copper and iron. Bishop Gerasim consecrated the main altar in the name of the Protection of the Holy Virgin. The left altar was dedicated to the Great Martyr Panteleimon, and the right to Saint Prince Alexander Nevsky.

On October 6, 1868, a church built with the funds of the residents of Buguruslan was consecrated in honor of the icon of the Mother of God "Joy of All Who Sorrow." This stone and warm church did not have a bell tower, and the bells were placed on wooden posts. In January 1877, the church suffered from a fire in the monastery, but it was soon restored and re-consecrated by the Samara bishop Gerasim on September 22 of the same year.

After the fire of 1877, a semi-stone house church in honor of the Nativity of John the Baptist was built, covered with shingles and painted with oil paint over plaster. It was consecrated on February 3, 1877, but was abolished after the opening of the Holy Trinity Church.

In 1893, the Samara bishop Guriy consecrated the site for the foundation of a new church. With the funds of merchant M.F. Rozhnov, a new warm stone three-altar church was built over six years. It had 17 domes with gilded iron crosses and was illuminated by 32 windows of various sizes. The consecration took place in 1899. The main altar was dedicated to the Holy Trinity. The left altar was consecrated in the name of the holy hierarchs Mitrophan of Voronezh, Tikhon of Zadonsk, Guriy of Kazan, and Theodosius of Chernigov. The right altar was dedicated to the icon of the Mother of God "Quick to Hear" and to the venerable Angelina of Serbia.
