The village of Lopukhivka was founded no later than the second half of the 18th century by the rivers Medveditsa and Itkarka. In 1817, a church was built here, after which the village was also called Nikolskoye. The stone building with a bell tower was erected by the efforts of the landowner Kineshsky. The chapel of the church was consecrated in honor of Saints Simeon the God-Receiver and Anna the Prophetess. The houses for the priest and the psalmist belonged to the church. The last owner of the village was landowner Yuryevich. Lopukhivka, located in the Atkarsky district of the Saratov province, was a volost center. In 1910, there were 1,006 residents in 166 households, and a zemstvo school and a feldsher station were operating.

During the Soviet era, the church of St. Nicholas the Bishop and Wonderworker lost its bell tower and was used as a grain storage facility.

In 2001, residents of the villages of Stara Lopukhivka, Belgaz, and Ozyornoye appealed to the Saratov diocese with a request to bless the restoration of the Holy Nicholas Church in Stara Lopukhivka. On February 7 of the same year, the first meeting of Orthodox believers took place, where members of the parish council were elected. In 2001, the church building received the status of a regional architectural monument.
