The church building was originally constructed for a Methodist congregation shortly after the end of the Civil War in the United States. In 1890, it was acquired by Lutherans and used until February 1919, when a group of Russian emigrants, numbering over 500, purchased it. The founders of the Holy Trinity Church were predominantly from areas of modern Belarus, as well as adjacent regions of Poland and Ukraine. Today, church services are conducted in accordance with Russian traditions.

Priest Vasily Kurdyumov was actively involved in establishing the parish, traveling from Philadelphia every Sunday to serve the Liturgy. By March 1919, the number of parishioners and the funds raised had increased so much that the parish was able to acquire its own church building. By American standards, this temple is considered old, as it was built in 1860.

The rector of the church, Father Ioann, recounted that services were then conducted entirely in Old Slavic, while communication was in Russian. All written records have been preserved, and they only show Russian names. Many of the founders lived long lives and attended the 75th anniversary celebration of the parish. Most parishioners were workers, with many men employed at the Port of Baltimore. In the 1920s and 1930s, shops began to open, and businessmen emerged among the parishioners. At that time, shops offered only two or three types of goods, and merchants would go door to door, offering meat products, dairy items, and fruits. Among these merchants were parishioners of the church. During World War II, some of them acquired special skills, became engineers, and worked in military factories, as Baltimore housed a large military base. Previously, a Russian school operated within the parish, where children studied the Russian language two to three times a month. Parishioners lived nearby and walked to the church.
