The wooden church was built in 1747 by Count Kirill Grigorievich Razumovsky and could accommodate up to 2000 parishioners. Surrounded by Old Believers and non-believers from the Polotnyany Plant, it served as a true fortress of Orthodoxy. The design of the new stone church in Karamyshevo was developed by junior architect N.P. Yushkov from the Kaluga provincial administration, and construction began in 1871. The new church with a bell tower was 25 fathoms long and 10 fathoms wide. The dome had seven large windows, and below, twenty. The stone church was completed in 1878 by nobleman Alexei Nikolayevich Larionov using church funds. In the refectory, chapels were arranged in honor of St. Basil the Great and St. Paraskeva Friday.

The church was closed in the 1930s, and the rector Ioann Meshchersky and the warden Pelageya Fedorovna Buylina were arrested. At a meeting of the troika of the OGPU in the Western region, Ioann was sentenced to three years of imprisonment, while Pelageya Fedorovna received three years of exile to the Northern region. The last rector, priest Pavel Volochkov, was sentenced on November 20, 1937, to 10 years in labor camps. During the Great Patriotic War, the bell tower and part of the refectory were destroyed, and later local residents dismantled the remaining parts for bricks.

In July 1999, resident B.V. Dudnikov from Kondrovo appealed to the diocese with a request to revive the church in Karamyshevo, attaching a petition from local residents, mainly from the village of Redkino. On September 30, 1999, a parish was established. On March 9, 2001, Archbishop Kliment of Kaluga and Borovsk appealed to the regional administration with a request to transfer the church to the diocese. Since October 8, 2001, priest Alexander Stukov has been the rector. Since 2003, a Sunday school has been operating at the church.
