The village of Ananyino is located 32 km southeast of the city of Barysh on the banks of the Syzran River. Previously, it was part of the Syzran district of the Simbirsk province. It is believed that the village was founded in 1700 by a serviceman from Penza, Luka Stepanovich Ananyin, who was granted 'wild land on the Mochaleyka and Poleyka rivers' covering an area of 350 quarters. Ananyino was settled by serfs from the Penza province. In the 1760s, the village passed to Nikolai Ivanovich Bakhmetyev, a corporal of the Semenovsky Life Guards Regiment, and after 1836 was divided between the Bakhmetyevs and the Sidelnikovs. In 1913, Ananyino had 131 households, 616 residents, a stone Sign Church, two chapels, and a church school. The well-known annual fair in the Simbirsk province took place in Ananyino from June 24 to 27 near the church. The feast day of the Sign was celebrated on December 10.

The stone three-altar church was built in 1818 with funds from landowner Maria Nikolaevna Maksimova. The church had three altars: the main one in honor of the Sign of the Mother of God, the right side altar in honor of the Tikhvin Icon of the Mother of God, and the left one in the name of Saint Nicholas the Wonderworker. The church housed the Tikhvin Icon of the Mother of God, revered as miraculous, which, according to church chronicles, repeatedly saved the peasant fields in the Karsun, Sengileev, and Syzran districts from drought and pests. A chapel was built at the site of the icon's appearance on the banks of the Syzran River.

Among the church's relics were silver eyes. It is said that a merchant, doubting the miraculous power of the icon, went blind, but after repentance and prayers, he regained his sight and, in memory of this, ordered two silver eyes, which were kept in the church.

The church, built according to the traditional 'octagon on a square' scheme, has an apse and a refectory with two side altars. To the west, a bell tower adjoined the refectory, of which two tiers of the square base have survived. The lower tier of the bell tower is pierced with high arched passages with wide archivolts and keystones. The church is topped with a low faceted dome on an octagonal base. The cross that crowned the church has been lost. The apse was covered with a conch, while the ceilings of the refectory and side altars have been lost. The first tier of the church, the apse, and the side altars have identical rectangular windows in shallow niches with sandriks. The doors of the church retain ancient wrought iron fittings, and iron grilles are inserted in the windows.
